Leishman stain, also known as Leishman's stain, is used in microscopy for staining blood smears. It is generally used to differentiate between and identify white blood cells, malaria parasites, and trypanosomas. It is based on a methanolic mixture of "polychromed" methylene blue (i.e. demethylated into various azures) and eosin. The methanolic stock solution is stable and also serves the purpose of directly fixing the smear eliminating a prefixing step. If a working solution is made by dilution with an aqueous buffer, the resulting mixture is very unstable and cannot be used for long.

Indigo carmine, or 5,5′-indigodisulfonic acid sodium salt, is an organic salt derived from indigo by sulfonation, which renders the compound soluble in water. It is approved for use as a food colorant in the U.S and E.U. It has the E number E132. It is also a pH indicator.

Product Application:
Indigo carmine in a 0.2% aqueous solution is blue at pH 11.4 and yellow at 13.0. Indigo carmine is also a redox indicator, turning yellow upon reduction. Another use is as a dissolved ozone indicator through the conversion to isatin-5-sulfonic acid. This reaction has been shown not to be specific to ozone, however: it also detects superoxide, an important distinction in cell physiology.It is also used as a dye in the manufacturing of capsules.
In obstetric surgery, indigo carmine solutions are sometimes employed to detect amniotic fluid leaks. In urologic surgery, intravenous injection of indigo carmine is often used to highlight portions of the urinary tract. The dye is filtered rapidly by the kidneys from the blood, and colors the urine blue. This enables structures of the urinary tract to be seen in the surgical field, and demonstrate if there is a leak. However, the dye can cause a potentially dangerous increase in blood pressure in some cases.

Sudan IV (C24H20N4O) is a lysochrome (fat-soluble dye) diazo dye used for the staining of lipids, triglycerides and lipoproteins on frozen paraffin sections. It has the appearance of reddish brown crystals with melting point 199 °C and maximum absorption at 520(357) nm.

Product Application:

• Sudan IV has been used industrially to color oils, waxes, shoe polish, greases, oily-resin lacquers, varnishes, cellulose acetate and acrylic resins, and in wood stains. As a biological stain, it is used primarily to stain lipids and fatty substances present in cells and tissues. The color imparted is a function of the solubility of the dye in the substance affected and does not involve chemical bonding. A saturated solution in isopropanol is prepared for use as a fat stain in animal tissue.
• Sudan IV is one of the dyes used for Sudan staining. Similar dyes include Oil Red O, Sudan III, and Sudan Black B. Staining is an important biochemical technique, offering the ability to visually qualify the presence of the fatty compound of interest without isolating it. For staining purposes, Sudan IV can be made up in propylene glycol. Alternatively, authors have reported using the dye saturated in isopropyl alcohol, 95% ethanol, or 0.05% by weight in acetone:ethanol:water (50:35:15). The idea is to use a moderately apolar solvent to solubilize the dye allowing it to partition into the highly apolar fat without the solvent solubilizing the fat to be stained.

Sudan III Stain, Saturated Alcoholic is used for identification of fat/lipid in frozen tissue sections. Sudan dyes are a group of fat/lipid soluble solvent dyes, also known as lysochromes. These solvent dyes readily stain fat/lipid due to the fact that the dyes are more soluble in lipid than in the solvents from which they are applied.
Sudan III is a lysochrome (fat-soluble dye) diazo dye. It is structurally related to azobenzene.

Product Application:
It is used to color nonpolar substances such as oils, fats, waxes, greases, various hydrocarbon products, and acrylic emulsions. Its main use is as a fuel dye in the United States of America mandated by the Internal Revenue Service (IRS) to distinguish low-taxed heating oil from automotive diesel fuel, and by the United States Environmental Protection Agency (EPA) to mark fuels with higher sulfur content; it is a replacement for Solvent Red 26 with better solubility in hydrocarbons
Sudan III is a dye used for Sudan staining. Similar dyes include Oil Red O, Sudan IV, and Sudan Black B. They are used for staining of triglycerides in frozen sections, and some protein bound lipids and lipoproteins on paraffin sections. It has the appearance of reddish brown crystals and a maximum absorption at 507(304) nm.

Sudan III is harmful if inhaled or swallowed, and can be absorbed through skin. MSDS rated a contact severity of 3, and cannot be made non-poisonous. Inhalation can cause intoxication and eye contact can lead to blurred vision and potentially blindness. The toxic effects exerted upon nervous system, particularly the optic nerve. Once absorbed into the body, it is very slowly eliminated. Symptoms of overexposure may include headache, drowsiness, nausea, vomiting, blurred vision, blindness, coma, and death.

Litmus, Powder is a mixture of dyes that are extracted from lichens and used to test whether materials are acidic or basic.
It is often adsorbed onto filter paper to produce one of the oldest forms of pH indicator, used to test materials for acidity.

Product Application:
The main use of litmus is to test whether a solution is acidic or basic. Blue litmus paper turns red under acidic conditions and red litmus paper turns blue under basic or alkaline conditions, with the color change occurring over the pH range 4.5–8.3 at 25 °C (77 °F). Neutral litmus paper is purple. Litmus can also be prepared as an aqueous solution that functions similarly. Under acidic conditions, the solution is red, and under alkaline conditions, the solution is blue.
